Innovative modular system for high-throughput protein purification

(I-Newswire) March 31, 2005 - The Vivapure� 8-to-96 well cobalt-chelate kit offers a unique modular format. Depending on the sample number, up to twelve individual 8-strips are inserted into frames with the footprint of standard 96-well plates. This feature results in proportionate savings when less than 96 samples are simultaneously processed.

Prior to protein purification, cell lysates are cleared by filtration through the multi-layer membrane of the Vivaclear� plus 8-strips. Afterwards, His-tagged proteins are isolated from the lysate using the Vivapure� cobalt-chelate 8-strips. Each well of these 8-strips is pre-packed with a membrane containing immobilized cobalt-IDA

(iminodiacetic acid) chelates, which specifically capture His-tagged proteins. Along with negligible unspecific protein binding, this membrane-based technology assures high and reproducible recoveries of ultrapure target proteins.

Parallel processing of multiple samples is accomplished by rapid centrifugation-based steps. Up to 250 �g His-tagged protein per well can be purified within 45 minutes. The kit is ideal for fast screening of expression libraries, optimisation of expression conditions and preparation of multiple purified proteins to be used in interaction studies, structural and functional analyses or for spotting protein arrays.

The Vivapure� 8-to-96 well cobalt-chelate kit enlarges the Vivapure family of membrane-based protein purification tools, which include a variety of centrifugal devices using metal chelate, ion exchange, C18 and Protein-A functional groups as well as the new kits for human serum albumin removal and adenovirus purification.
